I'm designing a 42" widescreen 16:9 touch screen interface. What resolution should I create my prototype in, what's a good resource site I should look into.
Regarding the resolution; I guess you're best of finding out what the hardware you're designing for runs as a native resolution and use that as a starting point. http://en.wikipedia.org/wiki/Display_resolution might be of some help.
When designing for touch-screen kiosks (for travel applications), I found the following publications useful:
